● 储存条件和效期:
该试剂盒常温贮存,常温运输。开封后有效期一年。
● 产品简介:
全血基因组DNA
小量提取试剂盒可从200-400 μl体积新鲜、冷冻或抗凝全血样本中快速分离纯化基因组DNA。该试剂盒可以在30分钟内完成单个或多个样品的操作。 提取过程无需使用蛋白酶K消化,不需要酚氯仿抽提,
也无需耗时的异丙醇沉淀等过程 。
使用该试剂盒提取到的DNA
可以用于PCR,Southern杂交,酶切消化等实验。
● 准备工作:
1. 准备无水乙醇;制冰机;1.5ml离心管;2ml离心管;离心机
2. 按照标签所示在去蛋白液GD和漂洗液PW中加入无水乙醇,混匀后盖紧瓶盖后常温贮存备用。
3. 每次使用前请检查缓冲液DL1和DL2是否有沉淀生成,如果出现沉淀,37℃温浴至沉淀溶解后再使用。
● 标准操作步骤:
如非指出,所有离心步骤均为使用台式离心机在常温下离心。
注意:以下方案适用于400
μl体积新鲜或冷冻血液样本。如果使用低于400 μl的全血样本,必须按比例减少缓冲液DL1和DL2的用量或者用1×PBS溶液补齐至400 μl,严格按照缓冲液DL1:全血:缓冲液DL2=3:4:3体积比进行操作,否则将导致后续操作无法进行。
1. 加入300μl缓冲液DL1于1.5
ml离心管中。
2. 加入400
μl抗凝全血,漩涡剧烈震荡30秒。 注:如果低于400 μl血液,用1×PBS补齐到400
μl。
3. 加入300μl缓冲液DL2,剧烈摇动离心管3-5次,漩涡剧烈震荡30秒-60秒。
注:加入缓冲液DL2后,会出现大量血红蛋白沉淀,必须剧烈震荡。
4. 12000 rpm 离心2分钟。
5. 把吸附柱装在2ml收集管中(已提供),将第4步得到的上清溶液全部转入柱中(小心别触及沉淀),12000
rpm离心1min,倒弃流出液重新套上收集管。
注:吸附柱的承受最大体积是700 μl,如上清超过此体积,可分2次离心,保证所有上清都加到柱中。
6.加入500 μl
去蛋白液GB(注意是否已经加入乙醇)至柱子中,12000 rpm离心1min,弃去流出液。
注:吸附柱膜上会有血色素残余,这是正常现象,可以被去蛋白液GD和漂洗液PW洗去。
7. 将吸附柱重新套回2ml收集管中,加入700μl 漂洗液PW(注意是否已经加入乙醇)至柱子中,12000 rpm离心1min,倒弃流出液;
8. 再加入500μl
漂洗液PW至柱子中,12000 rpm离心1min,
弃去流出液;
9. 将吸附柱重新套回2ml收集管中,12000 rpm离心空结合柱2 min以干燥柱子的基质;
注:这一步骤至关重要,不要省略。 否则残余乙醇会影响后续酶切或PCR实验。
10. 将柱子置于1.5ml灭菌离心管,加入50-80 μl65℃预热的洗脱缓冲液EB或灭菌去离子水至柱子的膜中央。 常温静置2 min;
注: ① 洗脱缓冲液体积最好不少于50 μl,体积过小影响回收效率。
② 洗脱液的pH值对于洗脱效率有很大影响。若用水做洗脱液应保证其pH值在7.0-8.5范围内,pH值低于7.0会降低洗脱效率。
11. 12000 rpm离心
2 min洗脱DNA。保留含DNA的流出液。将DNA储于-20℃。
● DNA浓度及纯度检测:
基因组DNA片段的大小与样品保存时间、操作过程中的剪切力等因素有关。提取的DNA片段可用琼脂糖凝胶电泳和紫外分光光度计检测浓度与纯度。可配制0.8-1.0%琼脂糖凝胶,使用λ/HindIII判断基因组的大小,完整的基因组大小应在23kb以上。使用分光光度计检测时, OD260/OD280比值应为1.7–1.9之间,如果洗脱时不使用洗脱缓冲液,而使用去离子水洗脱,比值可能偏低,但并不表明DNA纯度不高。
