● 产品简介:

    本产品是由7条带状双链DNA条带组成,适用于琼脂糖凝胶电泳中DNA条带的分析。产品中已含有1×loading buffer,可根据实验需要,直接取2-5 μl电泳,使用方便,电泳图像清晰。7条带的大小分别为250, 1000, 2500, 5000,7500,10000,15000bp。其中2500bp条带为100 ng/5μl,其余条带浓度为50 ng/5μl。

● 储存及运输:

4贮存3个月; -20长期贮存;常温运输。

● 使用方法:

1. 取2-5μl本产品加入到琼脂糖凝胶的加样孔中(每1 mm×1 mm加样孔加1 μl,如果加样孔宽于

5 mm,可以适当增加上样量)就行电泳。

2. 建议电泳条件:凝胶浓度为0.8-1.0%,凝胶长度10 cm,电泳电压~7v/cm,电泳时间50+分钟。

3. 通过核酸染料染色后紫外灯或蓝光仪下观察条带。

● 注意事项:

1. 琼脂糖的质量对DNA的电泳有很大影响,电泳时请尽量使用质量优等的琼脂糖。由于PAGE电泳的特殊性,该产品不建议用于PAGE电泳。

2. 使用与电泳缓冲液一致的缓冲液融化琼脂糖,例如使用1×TAE缓冲液电泳,需使用1×TAE缓冲液溶胶。

3. 多次电泳后缓冲液电导增强,相同电压下电流增大,导致分辨率降低,并且明显产热,严重时会引起凝胶熔化或DNA变性。

4. 推荐使用RealSafe系列染料(货号:GR002,GR003,GR004)染色。如果使用Goldview或EB等染料进行染色,由于此类染料带更多的正电荷(染料移动方向与核酸泳动方向相反),随着电泳时间延长,染料向大片段聚集,凝胶会出现阴阳胶现象(染料含量高的凝胶紫外下较亮,含量低的凝胶紫外下发暗),导致小分子量DNA显色很弱或不可见。阴阳胶可以在电泳结束后浸泡染色,将胶浸没在含1 μg /ml EB或Goldview的电泳缓冲液中20-25 min,小分子量DNA会显色清楚。更长时间浸泡会因DNA分子扩散而使条带弥散,小分子量DNA更易弥散。

5. 紫外照射下EB易分解,使DNA条带荧光变浅。DNA受紫外光照射形成嘧啶二聚体,不利于待回收片段的下游工作,因此尽可能减少紫外照射时间。

6. 蔗糖会结合DNA,降低其电泳迁移率,建议DNA样品上样使用含甘油的Loading Buffer。

7. 大部分核酸工具酶会结合DNA,降低其电泳迁移率,大分子量DNA尤为明显。
